Clinical studies have evaluated dapoxetine in men with premature ejaculation and concomitant erectile dysfunction who were already using a PDE5 inhibitor, showing improvement in ejaculation-related outcomes compared with placebo.
How It Works
Vardenafil works by enhancing the nitric oxide–cGMP pathway in penile tissue, which helps improve blood flow during sexual stimulation. Dapoxetine increases serotonin signaling by blocking serotonin reuptake, which can help delay ejaculation. The combination is intended to address both erection difficulty and premature ejaculation, although clinical benefit can vary between individuals.
Important Safety Information
Vardenafil should not be used with nitrates because of the risk of marked hypotension. Official prescribing information also warns about clinically important interactions with alpha-blockers and other blood-pressure-lowering medicines. Dapoxetine is a serotonergic medicine and should not be combined with MAO inhibitors; caution is also warranted with other serotonergic drugs because of the risk of serotonin syndrome. Possible Side Effects
Commonly reported adverse effects of vardenafil include headache, flushing, nasal congestion, and dyspepsia. Dapoxetine is commonly associated with nausea, dizziness, headache, and diarrhea. More serious reactions can include symptomatic low blood pressure, syncope, visual changes, priapism with PDE5 inhibitors, and serotonin syndrome with serotonergic combinations.
Scientific Evidence
Randomized clinical evidence shows that dapoxetine improves intravaginal ejaculatory latency time and patient-reported outcomes in premature ejaculation, including in men with concomitant erectile dysfunction already receiving a PDE5 inhibitor. A phase III study in such men found greater improvement with dapoxetine than placebo, with adverse events mainly reflecting known dapoxetine effects.
