How It Works
Naproxen reduces the formation of prostaglandins by inhibiting cyclo-oxygenase enzymes. Because prostaglandins contribute to pain, swelling and inflammation, lowering their production helps reduce these symptoms.
Important Safety Information
Naproxen is contraindicated in patients with hypersensitivity to naproxen or other NSAIDs, and in those who have had aspirin- or NSAID-triggered asthma, urticaria or allergic-type reactions. Regulatory information also warns against use in active or previous peptic ulcer disease, active gastrointestinal bleeding, severe renal, hepatic or heart failure, and during the last trimester of pregnancy. NSAIDs can increase the risk of serious cardiovascular thrombotic events and serious gastrointestinal bleeding, ulceration and perforation.
Clinically important interactions include other NSAIDs, aspirin, anticoagulants, antiplatelet medicines, corticosteroids, SSRIs, ACE inhibitors, ARBs and diuretics. Naproxen may also reduce the antiplatelet effect of low-dose aspirin when used together for more than one day.
Possible Side Effects
Common adverse effects include dyspepsia, abdominal pain, nausea, headache, rash, bruising and oedema. Important serious reactions include gastrointestinal bleeding, ulceration or perforation, hypersensitivity reactions including anaphylaxis, kidney problems, liver injury and severe skin reactions such as Stevens-Johnson syndrome, toxic epidermal necrolysis and DRESS.
Scientific Evidence
Human clinical evidence supports naproxen as an effective NSAID for pain and inflammation. Reviews and meta-analyses indicate that its analgesic and anti-inflammatory effects are well established, while safety concerns typical of the NSAID class remain important, especially for the gastrointestinal and cardiovascular systems. Some comparative analyses suggest naproxen may have a more favourable cardiovascular profile than certain other NSAIDs, but this does not remove class risks.
