How It Works
Vardenafil helps increase blood flow into the penis during sexual stimulation by inhibiting PDE5. This supports the normal erectile response, but it does not create an erection without sexual stimulation.
Important Safety Information
Vardenafil must not be used with nitrates or nitric oxide donors because the combination can cause a marked fall in blood pressure. Prescribing information also lists several important cardiovascular situations in which use is not recommended, including unstable angina, recent myocardial infarction or stroke, severe cardiac failure, hypotension, and uncontrolled hypertension. It should also be avoided in patients with severe hepatic impairment.
Clinically significant interactions are described with potent CYP3A4 inhibitors such as ketoconazole, itraconazole, ritonavir, indinavir, saquinavir, atazanavir, and clarithromycin, and with moderate CYP3A4 inhibitors such as erythromycin. Vardenafil should not be combined with other erectile dysfunction therapies unless specifically directed in official prescribing information.
Possible Side Effects
Common adverse effects reported with vardenafil include headache, flushing, nasal congestion, dyspepsia, dizziness, and back pain. More serious reactions can include significant blood pressure lowering, priapism, visual changes, hearing changes, and cardiovascular events in susceptible patients.
Scientific Evidence
Randomized controlled trials and review literature show that vardenafil improves erectile function scores and successful intercourse outcomes compared with placebo in men with erectile dysfunction. A 2021 systematic review and meta-analysis also supported its efficacy and tolerability across clinical trial data.
