How It Works
Tadalafil works by inhibiting PDE5, which helps increase blood flow in the penis during sexual stimulation. Dapoxetine works by increasing serotonin signalling in the nervous system, which can delay ejaculation. Together, the two medicines target different aspects of male sexual function.
Important Safety Information
Tadalafil must not be used with organic nitrates and is also contraindicated with riociguat. It can lower blood pressure, especially with alpha-blockers, antihypertensives or substantial alcohol intake. It should also be avoided in people for whom sexual activity is medically inadvisable because of cardiovascular status.
Dapoxetine should not be used with monoamine oxidase inhibitors, certain other serotonergic medicines, or in people with important cardiac rhythm problems, a history of fainting, or some psychiatric conditions such as mania or severe depression. Potent CYP3A4 inhibitors are also important interaction risks.
Because this is a combination product, safety concerns from both ingredients may apply together, including additive dizziness, faintness, and blood-pressure effects.
Possible Side Effects
Commonly reported adverse effects of dapoxetine include nausea, dizziness, headache, diarrhoea, and somnolence. Tadalafil is associated with headache, flushing, dyspepsia, nasal congestion, back pain and muscle pain, and it can also cause hypotension in susceptible patients. Rare but serious effects can include syncope, significant blood-pressure lowering, and severe hypersensitivity reactions.
Scientific Evidence
Randomized trials and meta-analyses show that dapoxetine improves premature ejaculation outcomes versus placebo, and tadalafil is effective for erectile dysfunction. Studies in men with both conditions suggest that combining dapoxetine with a PDE5 inhibitor can improve sexual outcomes, but it may also increase some adverse effects such as headache, flushing, and nasal congestion.
