How It Works
Tadalafil helps increase blood flow to the penis by inhibiting PDE5, which supports the erection response during sexual stimulation. Dapoxetine increases serotonin activity and can delay ejaculation. In combination, the two agents may help men who have both erectile dysfunction and premature ejaculation.
Important Safety Information
Tadalafil must not be used with nitrates because of the risk of a marked drop in blood pressure. It is also contraindicated with guanylate cyclase stimulators and should be used cautiously with alpha-blockers, antihypertensives, potent CYP3A4 inhibitors, and in people with significant cardiovascular, renal, or hepatic impairment. Dapoxetine carries important precautions related to serotonin syndrome, syncope, mood disorders, and interactions with other serotonergic medicines, monoamine oxidase inhibitors, thioridazine, and strong CYP3A4 inhibitors.
Possible Side Effects
Commonly reported effects of tadalafil include headache, flushing, dyspepsia, nasal congestion, back pain, and muscle pain. Dapoxetine commonly causes nausea, dizziness, headache, diarrhoea, and postural dizziness. Serious reactions can include fainting, prolonged erection, significant blood pressure changes, visual or hearing problems, and serotonin syndrome.
Scientific Evidence
Human studies have shown that dapoxetine improves premature ejaculation outcomes, and a randomized study in men with erectile dysfunction on PDE5 inhibitor therapy found benefit with dapoxetine. More recent clinical research comparing tadalafil, dapoxetine, and their combination reported greater improvement with the combination than with either treatment alone, although these findings do not establish that every fixed-dose product or strength has the same clinical profile.
