Description
Peppy (Sildenafil + Dapoxetine) 100 mg + 60 mg
Peppy is a combination medicine identified as containing sildenafil and dapoxetine. Based on the available regulatory and scientific sources, sildenafil is a phosphodiesterase type 5 (PDE5) inhibitor used for erectile dysfunction, while dapoxetine is a short-acting selective serotonin reuptake inhibitor (SSRI) studied and approved in some markets for premature ejaculation. The combination has been evaluated in clinical studies in men with premature ejaculation and concomitant erectile dysfunction.
Active Ingredient and Medicine Class
Sildenafil is a PDE5 inhibitor. This class helps increase penile blood flow during sexual stimulation. Dapoxetine is a selective serotonin reuptake inhibitor with rapid absorption and short action, developed for on-demand treatment of premature ejaculation.
Established Medical Uses
Sildenafil is established for erectile dysfunction in adult men. Dapoxetine is established in the treatment of premature ejaculation in men in jurisdictions where it is authorised. The fixed-dose combination has been studied in men who have both premature ejaculation and erectile dysfunction, but the exact regulatory status of the branded combination “Peppy” could not be confirmed from the sources reviewed.
How It Works
Sildenafil inhibits PDE5, which supports the natural erection process by enhancing the effects of nitric oxide and helping maintain higher levels of cyclic GMP in penile tissue. Dapoxetine increases serotonin signalling by inhibiting serotonin reuptake, which can delay ejaculation. The combination is therefore intended to address both erectile function and ejaculatory control.
Important Safety Information
Sildenafil must not be used with nitrates because of the risk of a marked fall in blood pressure. Caution is also needed with alpha-blockers and other blood-pressure-lowering medicines. Dapoxetine has important serotonergic interaction risks, including serotonin syndrome when combined with other serotonergic medicines. It is also associated with fainting, and caution is needed in people with significant heart disease or other conditions that may increase the risk of syncope. The combined use of sildenafil and dapoxetine has been studied, but the presence of two active agents means that safety concerns from both components remain relevant.
Possible Side Effects
Reported adverse effects include headache, dizziness, flushing, nausea, diarrhoea, somnolence, insomnia, and blood-pressure-related symptoms. More serious reactions can include syncope with dapoxetine, clinically significant hypotension with sildenafil, prolonged erection, and rare allergic or cardiovascular events.
Scientific Evidence
Peer-reviewed studies indicate that dapoxetine can improve intravaginal ejaculatory latency time and patient-reported control compared with placebo, with common but usually tolerable adverse effects. Studies of dapoxetine with sildenafil have shown that the two drugs do not produce a clinically important pharmacokinetic interaction, and clinical trials of the fixed combination have reported efficacy in men with both premature ejaculation and erectile dysfunction.
